Measuring AAV Quality Attributes with Light Scattering

Adeno-associated virus (AAV) is a common delivery vehicle in gene therapy. Robust, rapid and automated characterization methods are imperative for commercialization of AAV-based products. However, standard methods are labor- and time-intensive, requiring costly reagents. In this presentation, we will discuss a method to measure three critical quality attributes (CQAs) of AAVs:

  1. Total number of viral capsid particles;
  2. Relative capsid content (e.g., fraction of full capsids);
  3. Percentage of monomer or aggregates. The highly precise SEC-MALS method utilizes multi-angle light scattering in line with standard size-exclusion chromatography, requiring little sample prep, no reagents and minimal operator effort.
